まず understand を確実に土台に置くと覚えやすいです。意味は「理解する」。そこに mis- が付くと「誤って・うまくいかずに」の方向へずれるので、misunderstand で「誤って理解する」になります。miss とは別語ですが、感覚としては『理解がずれた』と捉えると定着しやすいです。人の発言・意図・状況を取り違える場面でよく使い、I misunderstood you. の形が頻出です。not understand が単なる『分からない』なのに対し、misunderstand は『違って受け取る』点が重要です。

misunderstandは古英語由来のmis-「誤って」+ understandan「理解する」から成る語です。understandは直訳の「下に立つ」ではなく「意味をしっかりつかむ」へ発達したため、全体で「つかみ違える」となります。understandやwithstandと同じstand系の語です。
Misunderstand is made from mis-, meaning "wrongly," and understand, which comes from Old English. Although understand looks like "stand under," the older sense developed into "grasp mentally" or "comprehend." So misunderstand came to mean "grasp wrongly" or "fail to understand correctly." It is related in form to other -stand words such as withstand.
